Different Methods of Chromatography Analysis General Introduction Health is of prime importance to a human beings and wants to get cured in the least possible time whenever they falls ill. This desire and necessity has resulted in the use of a large number of synthetic organic compounds as medicines despite the fact that usually side effects are associated with the use of these drugs. In recent times, practice of giving a number of drugs together has very much increased. Due to drug interaction, the levels of the active drugs together has very much increased. Due to drugs interaction, the levels of the active drug may be too high for a longer time to cause side effects. Further, the reduction/ oxidation products of these medicines, which are produced during the metabolism may also be responsible for their side effects. It is therefore necessary to develop sensitive trace analytical methods for the analysis of the drugs by using UV Spectrophotometer, most sophisticated and advanced chromatographic techniques like UPLC, GC, HPLC etc. Use of pharmaceutical preparations to make their determination, a matter of for most importance. Due to the great variability of the materials, which are to be analyzed; skillful sampling, preliminary clean-up procedure and selection of a appropriate method in the assay is necessary. With the introduction of new and more potent drugs every year, the pharmaceutical analyst is called upon to devise new analytical methods like chromatographic techniques; with ever increasing sensitivity, specificity and simplicity for new drugs . HISTORY OF CHROMATOGRAPHY1 The study of chromatography happening in eighteenth century when with a immense significance the nature of inorganic compounds was considered on filter paper by Runge. In 1898 Day in USA forced crude petroleum throughout a column of limestone and fullerââ¬â¢s earth. The chromatographic theory was discovered first by a Russian botanist , MichaelTswett (1906) who make use of a glass column of calcium carbonate for separation of chlorophyll pigments from plant by using petroleum ether. The major development occurred around 1930 when Lederer and co-workers in 1931 separated xanthine and lutein on a column of calcium carbonate powder. In 1935 ,Adams and Holmes observed some synthetic ion exchange resins capable of exchanging ionsà and thus ion exchange chromatography came in to existence. In 1944, Martin, Consden ,Gorden replaced silica gel columns by strips of filter paper and developed Paper chromatography. Thin layer chromatography though discovered first by Lzmailer and Shraiber , was further developed By Stahl and co-workers using silica gel on glass plates. Amongest the newest and most effective chromatographic technique for analyzing complex mixtures is Gas chromatography. It was introduced by Martin and James in 1952. INTRODUCTION OF CHROMATOGRAPHY1 A variety of methods are available for the separation of components from the mixture. They are mainly divided in to two types. Chemical methods Physical methods. The physical methods include fractional distillation, extraction, counter-current distribution; crystallization etc.These methods are effective in separation, purication and identification of many compounds .how ever difficulty arises in case of compounds where individual components have similar physical and chemical properties i.e mixture of liquids having very close boiling points etc. Chromatographic methods corresponds to the most handy and potent technique for these problems. These chromatographic methods are used for the partition of components of a composite mixture. Because of the quickness and efficiency of this methods, it can be used in all fields particulalarly in chemistry, biology, medicine, dyes, forensic departments and clinical studies. The term Chromatography derived from Greek words Kromatos means colour and Graphos ââ¬âmeans written. Tswett defined chromatography as the technique in which the components of a combination are separated on an adsorbent column in a flowing system. As per IUPAC Chromatography is defined as a method used mainly for the division of the components of a sample ,in which the components are disseminated between two phases, one of which is stationary whereas the other moves. The stationary phase might be a solid or a liquid support on a solid or a gel, and might be packed in a column ,spread as a layer or disseminated as a film. The mobile phase possibly will be gaseous or liquid. First and foremost for the partition of the components of a sample , in which the components are disseminated . TYPES OF CHROMATOGRAPHY: Chromatographic methods can be classified on the basis of stationary and mobile phases used, depending on the stationary and the mobile phase used, separation occurs because of a combination of two or more factors such as extent of adsorption, rate of migration and capillary action etcâ⬠¦ Different types of chromatographic techniques as follows a. Ion exchange chromatography A Adsorption Chromatography: The principle underlying the separation of the compounds is adsorption at the solid ââ¬âliquid interface, for successful separation , the compounds of a mixture must show different degrees of affinity for the solid support and the interaction between adsorbent and component must be reversible, as the adsorbent is washed with fresh solvent the various components will therefore move down the column until, ultimately, they are arranged in order of their affinity for the adsorbent ,those with least affinity move Paper chromatography: Paper partition chromatography was developed by consden et al, In this paper partition chromatography paper is used as the support or adsorbent but partition probably plays a greater part than adsorption in the separation of components of the mixtures In this chromatography substances are distributed between two liquids ie one is the stationary liquid (generally water )which is detained in the fibers of the paper and is called as stationary phase, the other is the touching liquid or rising solvent and is called moving phase, The components of the mixture to be separated at different rates and appear as spots at different points on the paper The moment of components on the paper depends on the amount and nature of the stationary phase compared with the amount of mobile phase in the same part of the paper and also on the partition coefficient The rate of movement of mobile phase at the solvent front tends to be faster than at the position of the component on the paper ,it is better to defined as Rf RF= DISTANCE traveled by centre of component /Distance travelled by solvent front Types of paper chromatography 1 Descending chromatography Is defined as while the development of the paper is made by permiting the solvent to travel down the paper Advantage: 1. The development can be continued indefinitely even though the solvents run off at the other end of the paper 2. Strong action exchange resins are prepared by sulphonating the free benzene rings. Strong anion exchange resins includes quaternary ammonium residues are prepared by chloromethylation of the free benzene rings followed by treatment with a tertiary amine salt ex: Trimethylamine amino hydrochloride. The strength and exchange capacities of ion exchange resins depend on the acidic or basic strength of the fine charged group. Thus the strongly acidic suphonic acid and strongly basic quaternary ammoinium groups give strong ion exchange resins with a high exchange capacity. Apparatus: the mode of operation of this system is isocratic i.e. one partition solvent or mixture is pumped throughout the analysis. For some determinations the solvent composition may be altered gradually to give gradient elution. Pumps: pumps are mandatory to distribute a stable flow of mobile phase at pressures varying from 1 to 550 bar. They are two types of pumps: Mechanical pumps: if the reciprocating piston type give a pulsating supply of mobile phase Dual piston reciprocating pump produce the two pistons are carefully phased so that simultaneously is filling the other is pumping. SUMMARY OF PRIDE AND PREJUDICE Pride and Prejudice is a story of Mr. and Mrs. Bennet and their five unmarried daughters---Jane, Elizabeth, Marry, Kitty, and Lydia. Mr. and Mrs. Bennet are desperate to see them all married. News is that a wealthy young gentleman named Charles Bingley has rented the estate of Netherfield Park near the Bennet estate. Mr. Bennet decides to make a special visit to Mr. Bingley to talk about his eldest daughter Jane. The Bennets attend a ball where Mr. Bingley is present and is taken by Jane and spends the whole night dancing with her. Mr. Bingley brought with him his friend, Mr. Darcy. Mr. Darcy is arrogant and not very pleased with the evening and out of arrogance, refuses to dance with Elizabeth. At many social events over an amount of weeks, however, Mr. Darcy finds himself more and more attracted to Elizabeth. Jane?s friendship with Mr. Bingley also continues to grow, and Jane finds herself going to visit Mr. Bingley at his mansion. On her way to the estate she gets caught in the rain and gets very ill forcing her to stay for many days. In order for Elizabeth to go get Jane she has to hike through the fields getting her clothes dirty. When she arrives to the estate, Miss Bingley notices that Darcy, whom she likes, pays quite a bit of attention to Elizabeth. When Jane and Elizabeth return home, Mr. Collins, a young clergyman who stands to inherit Mr. Bennet?s property, is there visiting their household. During his stay with the Bennet?s, he makes a marriage proposal to Elizabeth. She turns his offer down and injures his pride. Meanwhile, the Bennet girls have made friends with the military officers stationed nearby. One of those military officers is Wickham, a handsome young soldier who likes Elizabeth and tells her how Darcy cheated him out of an inheritance. Disappointing Jane, the Bingleys and Mr. Darcy left their estate and returned to London at the beginning of winter. Furthermore, Elizabeth receives appalling news that her best friend Charlotte Lucas is engaged to Mr. Collins. Charlotte explains to Elizabeth that she needs the match for financial reasons and marries Mr. Collins. As winter continues, Jane visits the city in hope to see Mr. Bingley instead Miss Bingley visits her first. While Miss Bingley?s visit was very abrupt, Mr. Bingley never shows up. The marriage search for the Bennet girls appears hopeless.

My Beloved Charioteer Essay
Shashi Deshpande is an award-winning Indian novelist. Shashi Deshpande makes gender central to her writing. In her story ââ¬Å"MY BELOVED CHARIOTEERâ⬠she tries to the show the relationships of grandmother, mother and daughter at various stages of life. It is a story of a mother daughter relationship as well as a womenââ¬â¢s role as a wife. Mother and daughter relationship is like sisters or sparring partners. They care for each other. My Beloved Charioteer depicts the life of motherââ¬â¢s nature and behavior not as generosity but as a normal human being full of negatives as well as positives, full of emotions as well as frustrations and not a perfect being. My Beloved Charioteer by Shashi Deshpande is based on the three generations, three women i.e. grandmother, mother and daughter and three different mindsets living together. Grandmother being the oldest one and the one who takes care of each and everyone in the house. Mother named Arti the daughter of grandmother a depressed lady as for whom she cared and loved have died. She loved two persons that is her father and her husband has both died. She is now the most irritated person in the house and she spends her most of the time smoking and looking at the walls of her room. She doesnââ¬â¢t talk to anyone neither to her daughter Priti and nor to her mother. She evens scolds priti for no reason .She is so much entangled with the past that her present is neglected. The daughter named priti the youngest and the most energetic character. Her father and grandfather had both died. Her mother doesnââ¬â¢t talk to her. Her caretaker and her best friend in the house is her grandmother (Ajji). When Priti is gone silence settles in the house. The relation between the Arti and Priti is not too good in the story. Arti is depressed, sad, unhappy, and is in pain. She doesnââ¬â¢t do any work, which a mother should do for her daughter and for her aged mother. She doesnââ¬â¢t talk to Priti; play with her however she scolds Priti at times to show her anger, outrage and displeasure. Because of her nature grandmother Ajji have to take care of Priti, she acts like her mother. She wakes her, she dresses her for school, make breakfast for her. It is shocking to seeà that at this age also Ajji does all the work and she even doesnââ¬â¢t complain. The relationship of Arti and Priti is not good whereas the relationship between Ajji and Priti is good. Ajji once insist Arti to go out of house like park whereas in reply she says that seeing other people happy she wanted to kill and bury their happiness. This shows one of the natures of depressed lady. Ironically Ajjiââ¬â¢s husbandââ¬â¢s room offers her the opportunity to find her voice, rebel against her daughter and break the silence that threatens to destroy her home and her granddaughter. The photo frame that was broken accidently by Arti gave a chance, which helped Ajji to talk to her and to explain her that what gone is gone. Ajji is also holding the pain of her husband but on the other hand she is managing and not running from her problems and fears. Priti has a daughter and she need to take care of her. Ajji also says that she does not want to ever see the photograph again as it is no point of seeing things which gives pain and discomfort. Arti needs to be happy at which she got rather than things, which she doesnââ¬â¢t have. Hence through this story Shashi Deshpande elegantly brings out the mother/daughter relationship on different stages and aspects of life.

Global Warming is one of the biggest problems in human history and the destiny of our planet is in our hands. If nothing is done, horrible things will happen, for example, Antarctica's animal population is decreasing. Also, the environment is changing because of constant temperature changes caused by the greenhouse effect. Antarctica's animal population is moving and ending their hibernation's early. The polar bears are thinner and less healthy than those of 20 years ago. Many of fish are moving northward to find colder water, so that they can survive and produce offspring. This is happening because the rays of the sun get through the ozone layer melting the icecaps making the water heat up. The marmots end their hibernation three weeks earlier than usual because of the warm weather. We should care because if all these things are happening to the animalââ¬â¢s maybe they will eventually happen to us. I am also concerned about the environment changing because of constant temperature change, caused by the Greenhouse Effect. The Greenhouse Effect is important. Without the Greenhouse effect, the Earth would not be warm enough for humans to live. But if the Greenhouse effect becomes stronger, it could make the Earth warmer than usual. The American Civil War was a conflict between the Union and the Confederacy. It was a war between the states. The North was fighting to abolish slavery and the South was fighting for their independence. During the war, roughly three million people fought for their cause. The majority of these three million soldiers are sung heroes. However, there are still those heros who people are unaware of. One rarely hears about these unsung heroes who were of tremendous assistance to the cause. These heroes are the southern slaves who became soldiers and joined the Union Army. Although many do not know of their role, the black soldiers played a very significant part in the American Civil war. There were many different black regiments that took part in the fight for freedom. Some examples of such regiments were the 54th Massachusetts, the Black Brigade of Cincinnatti, and the First Regiment South Carolina Volunteers. In addition to these, there were also countless others. Hundreds of thousands of black men wanted the chance to fight for their freedom. The 54th Massachusetts was organized in March of 1863 at Camp Meigs, Readville, Massachusetts by Robert Gould Shaw. This was one of the first all black regiments formed. Free blacks from the north, particularly Massachusetts and Pennsylvania, made up this regiment. The main sacrifice made was life. Soon after being formed, the 54th engaged in battle at Battery Wagner. There was over 250 men killed. Included was the founder Colonel Shaw, who died on the crest of the enemy parapet shouting, ââ¬Å"Forward, Fifty-fourth!â⬠2 Several other members of the 54th either risked or lost their lives. One such person was Sgt.! William H. Carney. He received the Medal of Honor for risking his life in action. His actions were described as follows: ââ¬Å"When the color sergeant was shot down, this soldier grasped the flag, led the way to the parapet, and planted the colors thereon. When the troo... But both sides are adamant on wanting the complete land which I believe is not ever possible. The result will be that the blood shed will continue on the both sides. Until and unless both the communities do not talk on the humanitarian basis this, all will remain for the next generations to suffer.
